Resumo Grande parte dos solos agricultáveis brasileiros apresenta deficiência de P, em função da maior parte deste elemento estar indisponível para a absorção pelas plantas. Neste contexto, a habilidade dos fungos filamentosos em solubilizar o P de várias fontes, sejam estas, insolúveis, orgânicas e/ou inorgânicas, tem sido considerada uma característica de destacada importância. O objetivo do presente estudo foi caracterizar a micobiota produtora de fitase no Bioma Cerrado, bem como, apresentar uma caracterização dos fatores abióticos do presente bioma e as possíveis correlações com a micobiota encontrada. As amostras foram coletadas de oito fragmentos florestais, em duas épocas, duas profundidades do solo (0-5 e 5-10 cm) e na camada de serapilheira para a quantificação da micobiota e avaliação da produção de fitase. As coletas foram realizadas em duas épocas, sendo uma na época chuvosa e outra na época seca. Na coleta da época chuvosa, o número total de fungos produtores de fitase foi menor, mas o número de Aspergillus seção Nigri produtores de fitase foi maior. Na época seca, o número de fungos produtores de fitase pertencentes ao gênero Penicillium foi maior. O fator época do ano alterou qualitativamente e quantitativamente a micobiota produtora de fitase. Concluiu-se que os solos do Bioma Cerrado são potencialmente exploráveis para a obtenção de fungos solubilizadores de P, sendo a época seca do ano a mais propícia para este fim.

Abstract The majority of Brazilian arable soils shows P deficiency, due to the fact that most of this element in the soil is unavailable for the plant absorption, for this and other reasons, the ability of filamentous fungi to solubilize P from various sources, insoluble, organic and inorganic, has been considered a desirable feature for several uses. The aim of this study was to characterize the production of phytase mycobiota in soil located in Cerrado in two seasons, under dry and rainy seasons, and also characterize abiotic factors in this biome in order to verify the existence of correlations between the quantitative and qualitative aspects of their mycobiotas in these conditions. The samples were collected from eight forest fragments twice, two soil depths (0-5 and 5-10 cm) and litter layer to quantify mycobiota and the evaluation of phytase production. In the collection carried out in the rainy season, the total number of phytase producing fungi was lower, but the number of Aspergillus section Nigri producers of phytase was greater. In the dry season the number of phytase producing fungi belonging to the genus Penicillium was higher. The year season´s factor alters quantitatively and qualitatively the phytase producer mycobiota. It was concluded that the soils are potentially exploitable in relation to obtaining phosphorus solubilizing fungi, and the dry season is more propitiate to the achievement of these fungi.

RESUMO: A utilização dos fungos como fonte de enzimas vem adquirindo status de destaque nas mais variadas áreas industriais e comerciais, e os Aspergillus membros da seção Nigri possuem significativo potencial para produção de enzimas. Os objetivos deste estudo foram isolar e avaliar Aspergillus da seção Nigri de serrapilheira e solos do bioma Mata Atlântica quanto à produção de enzimas hidrolíticas. Os ensaios para produção das enzimas foram realizados em placas de Petri, utilizando diferentes meios de cultivo, adequados ao crescimento microbiano e com a presença dos respectivos substratos indutores à produção das enzimas - celulases (carboximetilcelulose), proteases (leite desnatado), amilases (amido solúvel), pectinases (pectina cítrica) e fitase (meio Pikovskaya). Foram isolados 42 fungos, sendo desse total 16,7% provenientes da camada de serrapilheira e 83,3% provenientes do solo na profundidade de 0 a 5 cm e 5 a 10 cm. Todas as linhagens isoladas apresentaram produção de amilases, protease e fitase, 90,4% produziram celulase, e nenhuma linhagem produziu pectinase. Com esses resultados, percebeu-se significativo potencial dos fungos da Mata Atlântica como produtores de enzimas hidrolíticas. As avaliações da atividade enzimática apresentaram resultado satisfatório quando comparados à literatura científica.

ABSTRACT: The use of fungi as a source of enzymes has become widespread in various industrial and commercial areas, and Aspergillus section Nigri has significant potential for producing enzymes. The aim of this study was to isolate Aspergillus section Nigri from plant litter and soil from the Atlantic Forest biome and evaluate it with regards to hydrolytic enzyme production. The trials for producing the enzymes were carried out in Petri dishes, using different culture mediums adapted for microbial growth and with the respective substrates for inducing enzyme production - cellulase (carboxymethyl cellulose), protease (skimmed milk), amylase (soluble starch), pectinase (citrus pectin), and phytase (Pikovskaya medium). Forty-two fungi were isolated, 16.7% derived from the plant litter layer and 83.3% derived from soil at a depth of 0 to 5 cm and 5 to 10 cm. All of the isolated lineages presented amylase, protease, and phytase production, with 90.4% producing cellulase and no lineage producing pectinase. From the results, the significant potential for Atlantic Forest fungi as hydrolytic enzyme producers could be perceived. The enzymatic activity evaluations presented a satisfactory result when compared with the scientific literature.

RESUMO: Os fungos entomopatogênicos são importantes agentes de controle biológico em todo o mundo e têm sido objeto de intensa pesquisa por mais de 100 anos, infectando artrópodes na natureza e podendo ocorrer em níveis enzoóticos ou epizoóticos em suas populações de hospedeiros. O seu mecanismo de infecção envolve a fixação do esporo à cutícula do inseto, seguido da germinação, penetração da cutícula e disseminação interna no inseto. As linhagens dos fungos entomopatogênicos estão concentradas nas ordens: Hypocreales (vários gêneros), Onygenales (gênero Ascosphaera), Entomophthorales e Neozygitales (Entomophthoromycota).

ABSTRACT: Entomopathogenic fungi are important biological control agents throughout the world, have been the subject of intensive research for more than 100 years, and can occur at epizootic or enzootic levels in their host populations. Their mode of action against insects involves attaching a spore to the insect cuticle, followed by germination, penetration of the cuticle, and dissemination inside the insect. Strains of entomopathogenic fungi are concentrated in the following orders: Hypocreales (various genera), Onygenales (Ascosphaera genus), Entomophthorales, and Neozygitales (Entomophthoromycota).

Aflatoxinas são substâncias carcinogênicas de ocorrência natural, extremamente tóxicas para humanos, que têm sido identificadas no trigo e em produtos derivados. O uso de métodos analíticos confiáveis para avaliar e monitorar tais contaminantes é de grande importância. Este trabalho objetivou validar, intralaboratorialmente, uma metodologia para a determinação de aflatoxinas B1, B2, G1 e G2, em grãos de trigo, utilizando derivatização pré-coluna e quantificação por Cromatografia Líquida de Alta Eficiência com detecção por fluorescência (CLAE-DF). Três metodologias foram avaliadas e a que se adequou para tal propósito baseou-se na extração com clorofórmio, remoção dos interferentes por filtração, partição líquido-líquido com metanol-água-hexano e metanol-água-clorofórmio e derivatização pré-coluna com ácido trifluoracético. O método apresentou variação de repetibilidade inferior a 15% e recuperação na faixa de 70-110%, com limites de detecção e quantificação (0,6 µg kg-1 e 1,2 µg kg-1, respectivamente) inferiores ao nível máximo permitido pela Comissão Europeia (4,0 µg kg-1) e pela legislação brasileira (5,0 µg kg-1), para a presença de aflatoxinas em trigo e derivados. Por meio desta metodologia, foram quantificadas aflatoxinas em 20 amostras comerciais de grãos, farelo de trigo e farinha de trigo integral e refinada, destinadas ao consumo humano direto. Seis amostras (30%) foram positivas para aflatoxinas e todas apresentaram-se dentro do limite regulatório permitido pela legislação brasileira.

Aflatoxins are naturally occurring carcinogenic substances, extremely toxic to humans, which have been identified in wheat and wheat by-products. The use of reliable analytical methods to evaluate and monitor such contaminants is extremely important. This study aimed to in-house validate a methodology to determine aflatoxins B1, B2, G1 and G2 in wheat grains using pre-column derivatization and quantification by High Performance Liquid Chromatography with fluorescence detection (HPLC-FLD). Three methods were evaluated and the most suitable one was the method based on the extraction with chloroform, removal of interfering chemicals by filtration, liquid-liquid partition with hexane-methanol-water and methanol-water-chloroform and pre-column derivatization with trifluoroacetic acid. The method showed a Relative Standard Deviation lower than 15% and recovery values in the 70-110% range, with limits of detection and quantification (0.6 µg kg-1 and 1.2 µg kg-1, respectively) below the maximum level of aflatoxins allowed in wheat and wheat by-products by the European Commission (4.0 µg kg-1) and by the Brazilian legislation (5.0 µg kg-1). Using the validated method, aflatoxins were quantified in 20 commercial samples of wheat grains, wheat bran, whole wheat flour and refined wheat flour intended for direct human consumption. Six samples (30%) were positive for aflatoxins and all samples presented levels below the maximum limit stipulated by the Brazilian legislation.

This study aimed to determine the occurrence of AFM1 contamination in the samples of grated parmesan cheese marketed in the Metropolitan Region of Rio de Janeiro -Brazil. Thirty samples representing 10 major brands marketed in the region were analyzed by High Performance Liquid Chromatography with fluorescence detection (HPLC-FLD) after purification with immunoaffinity column. The method showed recovery values within the range of 70-90%, with RSD lower than 15% and limits of detection and quantification below the maximum level allowed by the European Commission for the presence of AFM1 in cheeses. The mycotoxin was identified in 18 (60%) of the grated cheese samples tested. The highest value corresponded to 0.69 ± 0.02 µg/kg and the mean for all the analyzed samples was 0.16 µg/kg. All the samples were lower than the limit established by the Brazilian legislation (2.5 µg/kg) for AFM1 in cheeses in general. However, eight samples (26.7%) presented AFM1 levels above the tolerance limit of 0.25 µg/kg adopted by the European Commission. These results indicated that AFM1 levels in the grated cheese consumed in Rio de Janeiro -Brazil were relatively high and it could provide a potential hazard for the public health.

O presente trabalho teve como objetivo fornecer dados para uma melhor compreensão entre a interação de microrganismos, flora e solo. O estudo foi realizado no Parque Natural Municipal do Curió, Paracambi, RJ. Foram selecionadas duas áreas, com diferentes graus de alteração antrópica, sendo em cada uma destas delimitado um talhão de aproximadamente 1.000 m², e nestes foram realizados levantamentos floristicos e coletas de amostras de terra e serrapilheira para a avaliação da fertilidade e dos microrganismos. Nas duas áreas quantificou-se um total de 89 espécies, reunidas em 59 gêneros e 33 famílias, entre espécies arbóreas, arbustivas, herbáceas e trepadeiras. Na área com maior grau de antropização ocorreram os maiores valores de pH, Ca e K, indicando um ambiente de maior eutrofismo. Já na área com menor grau de alteração, observaram-se maiores valores de H + Al, decorrentes da maior quantidade de material orgânico. Para a serrapilheira, foi verificado um padrão com tendência similar a observada para o solo. Os maiores valores de serrapilheira aportada, também contribuem para os maiores valores de K, elemento facilmente lixiviável no material em decomposição. Quanto à abundância dos microrganismos tanto na serrapilheira como no solo foi observada uma distribuição diferenciada dos microrganismos devido a variação da umidade relativa e temperatura, sendo as bactérias as predominantes, padrão este também observado para as diferentes áreas.

This study aimed to better understand the interaction between microorganisms, plants and soil. The study was conducted in Parque Natural Municipal do Curió (Paracambi, RJ). We selected two areas with different degrees of human disturbance, and in each of these enclosed a plot of 1,000 m². Floristic surveys were carried out and litter and soil samples were collected to evaluate fertility and the microorganisms. In both areas, 89 species were found, within 59 genera and 33 families, of trees, shrubs, herbs and vines. The highest values of pH, Ca and K were found in the area with the highest degree of human disturbance, indicating an environment of increased tropism. In the area with a lower degree of alteration, we observed higher values of H + Al, resulting from a higher amount of organic material. For the litter, the pattern was similar to that found for the soil. The highest values for the leaf litter also contributed to higher values of K, which is an element that easily leaches from mulch. Considering the abundance of microorganisms in both the litter and in soil, an uneven distribution of microorganisms was found due to the variation of relative humidity and temperature; bacteria were predominant, a pattern observed for both areas.

O presente trabalho teve como objetivo avaliar a diversidade e a sucessão da família Trichocomaceae em áreas com diferentes graus de antropização, denominadas área mais impactada e área menos impactada. O estudo foi realizado no Parque Natural Municipal do Curió, Paracambi-RJ. Foram selecionadas duas áreas, com diferentes graus de alteração antrópica, sendo em cada uma destas delimitado um talhão de aproximadamente 1.000 m², sendo nestes realizadas coletas de terra e serrapilheira para a avaliação da fertilidade e da micobiota. De maneira geral, observa-se que, na área mais impactada, ocorreram os maiores valores de pH, Ca e K. Os maiores valores de serrapilheira aportada, observados nesta área, também contribuem para os maiores valores de K, elemento facilmente lixiviável no material em decomposição. Os maiores valores de Unidades Formadoras de Colônias (UFC) no solo e na serrapilheira, 9 × 10(4) e 5,4 × 10(4), respectivamente, foram observados na área mais impactada. Também nesta área foram verificados os maiores valores de Ca no solo e na serrapilheira, o que sugere que esse nutriente pode estar contribuindo para o maior número de UFCs. Para a área menos impactada, os valores de UFC estiveram entre 4 × 10(4) para serrapilheira e 5 × 10(4) para o solo, não sendo observada nenhuma associação clara entre um maior número de UFCs e os teores de nutrientes no solo e/ou na serrapilheira. O número total de fungos isolados foi de 87, pertencentes a quatro gêneros e 22 espécies diferentes, sendo 16 Penicillium spp., quatro Aspergillus spp., um Paecilomyices sp. e um Trichoderma sp. Não foi observada uma clara associação entre as variáveis climáticas e o número de UFCs. O maior número de UFCs foi observado na área mais impactada tanto no solo quanto na serrapilheira. Os maiores teores de Ca quantificados nesta área sugerem que esse nutriente pode estar contribuindo para o aumento das UFCs. O genêro Penicillum, em ambas as áreas, foi o mais representativo.

The purpose of the present study was to assess the diversity and succession of family Trichocomaceae in areas with different levels of human disturbance, namely, most impacted area and least impacted area. This research was carried out in "Parque Natural Municipal Curió" (Natural City Preserve) in Paracambi, State of Rio de Janeiro. Two areas with different levels of human disturbance were selected and, in each of these areas, a 1,000 m² plot was delimited, where soil and leaf litter samplings were collected for fertility and mycobiota assessment. In general, the most impacted area presented the highest values for pH, Ca and K. The highest values of leaf litter yield were also observed in this area, which contributed to the higher values of K - an element easily leachable in mulch. The highest values of Colony Formation Units (CFU) in soil and litter: 5.4 × 10(4) and 9 × 10(4), respectively, were observed in the most impacted area. This area also presented the highest values for Ca in soil and litter, which suggests that this nutrient may contribute to the greater number of CFUs. For the least impacted area, the CFU values were between 4 × 10(4) for litter and 5 × 10(4) for soil; no clear association was observed between the higher number of CFUs and the concentrations of nutrients in soil and/or litter. The total number of fungi isolated was 87, belonging to four different genera and 22 species, with 16 Penicillium spp., four Aspergillus spp., one Paecilomyces sp. and one Trichoderma sp. No clear association was noticed between climatic variables and the number of CFUs. The greatest amount of CFUs was observed in the most impacted area, both in soil and leaf litter. The higher concentration of Ca quantified in this area suggests that this nutrient may be contributing to the increase in CFUs. The genera Penicillium spp. was the most representative fungus in both areas.

The aim of the present work was to select filamentous fungi isolated from diverse substrates to obtain the strains with potential to produce the hydrolytic enzymes. From a total of 215 strains, seven strains from the soils, six from the plants and one from sugarcane bagasse were selected and identified as belonging to the Trichoderma, Penicillium and Aspergillus genera. The best hydrolytic activities obtained by semi-solid fermentation using these strains were approximately: 35; 1; 160; 170 and 120 U/gdm (CMCase, FPase, β-glucosidase, xylanase and polygalacturonase, respectively), demonstrating their potential to synthesize the enzymes compared with the results reported in the literature.

As formigas do gênero Atta são pragas importantes de diversas culturas agrícolas, pastagens e reflorestamentos. Os fungos entomopatogênicos estão entre os fatores naturais de mortalidade dessas formigas e por isso apresentam potencial para serem usados no controle biológico dessa praga. O presente trabalho teve como objetivo selecionar isolados de Metarhizium anisopliae e Beauveria bassiana patogênicos a soldados de Atta bisphaerica e Atta sexdens rubropilosa em condições de laboratório. Para a avaliação da patogenicidade, foram utilizados oito isolados de M. anisopliae e seis de B. bassiana. O experimento foi conduzido em DIC, sendo a parcela composta por um grupo de 10 soldados por espécie de formiga, sendo utilizadas três repetições por tratamento. Para cada isolado, três grupos de soldados foram pulverizados com suspensão de 1,0 x 10(8) conídios ml-1 e mantidos em câmara úmida (25±1°C, 80±1% de UR e no escuro) sem alimentação, sendo a mortalidade verificada diariamente. Dos 14 isolados testados, quatro de M. anisopliae e quatro de B. bassiana foram patogênicos aos soldados de ambas as espécies de formigas. A virulência foi avaliada para os isolados que causaram mortalidade igual ou maior a 50%. Para cada isolado, suspensões contendo 1,0 x 10(6) a 1,0 x 10(11) conídios ml-1 foram pulverizadas sobre três grupos de 10 soldados e igualmente acondicionados como no teste de patogenicidade. A porcentagem de mortalidade foi calculada a cada 24 horas para determinação do TL50. O isolado ENA04 de M. anisopliae foi mais patogênico, causando mais de 80% de mortalidade nos primeiros três dias após a inoculação, apresentou maior capacidade de esporular nos cadáveres dos soldados e foi o mais virulento para os soldados de A. bisphaerica, com um TL50 de 1,15 dias. Todos os isolados patogênicos aos soldados de A. sexdens rubropilosa foram igualmente virulentos.

The ants of the genus Atta are important pests of several crops, pastures and planted forests. The entomopathogenic fungi are among the natural mortality factors of these ants and because of that they have potential to be use in biological control of this pest. The present research aimed to select isolates of Metarhizium anisopliae and Beauveria bassiana pathogenic to soldiers of Atta sexdens rubropilosa and Atta bisphaerica under laboratory conditions. To evaluate the pathogenicity, eight isolates of M. anisopliae and six of B. bassiana were used. The experiment was carried out in a completely randomized design, being the plots composed by a group of ten soldiers per each ant species, and three replicates per treatment. For each isolate, three groups of soldiers were sprayed with conidial suspensions containing 1.0 x 10(8) conidia ml-1, kept in moist chamber (25±1°C, 80 1% of RH in the dark) without food, and the mortality was evaluated every day. From the 14 isolates tested, four of M. anisopliae and four of B. bassiana were pathogenic to soldiers of the both ant species. The virulence was evaluated to the isolates that caused mortality equal or higher than 50%. For each isolate, suspensions containing 1.0 x 10(6) to 1.0 x 10(11) conidia ml-1 were sprayed on three groups of ten soldiers, and equally kept as on the pathogenicity test. The mortality percentage was calculated each 24 hours for determining TL50. The isolate ENA04 of M. anisopliae was the most pathogenic, causing more than 80% of mortality in the first three days after inoculation, showed higher capacity of spore production on ant cadavers, and was the most virulent to the soldiers of A. bisphaerica, with a TL50 of 1.15 days. All pathogenic isolates of A. sexdens rubropilosa soldiers were equally virulent.

RESUMO A família Trichocomaceae inclui os gêneros de fungos anamórficos, Aspergillus e Penicillium. Muitos membros dessa família são importantes causadores de degradação de alimentos, biodeterioração, patogênicos a animais, e algumas espécies são usadas em biotecnologia e podem produzir micotoxinas. O presente trabalho foi realizado em duas áreas, durante o período de um ano (2006-2007) com coletas em área de plantio de pinus (Pinus elliottii) e Corymbia (Corymbia citriodora) com idade aproximada de 20 anos, no campus da UFRRJ, Seropédica, Rio de Janeiro, Brasil. Na área de cada cobertura vegetal foi estabelecida uma parcela de aproximadamente 500 m2, onde foram coletadas aleatoriamente dez amostras simples, em intervalos regulares de 70 dias. No momento da coleta foram medidas as temperaturas do solo com geotermômetro digital. Durante o de estudo também foi quantificada a precipitação pluviométrica e a temperatura ambiente. No período, foi verificada uma pequena variação da temperatura do solo nas áreas, valores de 21,93 a 27,69oC na área de pinus e 22,22 a 26,58oC na área de Corymbia. A variação mensal da umidade relativa mínima foi de 27,2/20,5 e máxima de 82,6/63,2 e os maiores valores de precipitação observados foi no mês de janeiro (22 dias). Em relação à micobiota, foi observada uma variação crescente da unidade formadora de colônia (UFC) nos período de maior temperatura e umidade relativa com índices variando de 12,8/58,2 e 20,3/83,3x103 para Corymbia e pinus respectivamente. O número total de fungos foi de 190 isolados, pertencentes a cinco gêneros e 54 espécies diferente, sendo 32 Penicillium spp., 19 Aspergillus spp., um Eupenicillium javanicu, um Eurotium chevalieri e um Sclerocleista ornata. A espécie mais abundante foi Penicillium decumbens, sendo encontrada em todas as coletas. Os períodos de maior concentração de UFC estão correlacionados com os períodos de chuva, umidade relativa e temperatura. As variáveis climáticas precipitação e temperatura foram os fatores que mais influenciaram na sucessão da micobiota no solo.

ABSTRACT Trichocomaceae family encompasses the greatest anamorphic fungi genera: Aspergillus and Penicillium. Many from this family have been causing food degradation, biodeterioration, animal pathologies, and some species have also been used in biotechnology as well as being responsible for mycotoxins production. This survey was carried out from 2006 to 2007 on approximately 20 years old Pinus (Pinus elliottii) and Corymbia (Corymbia citriodora) two plantation areas at Universidade Federal Rural do Rio de Janeiro, Rio de Janeiro, Brazil. At each litter area, around 500 m2, parcel was set up where 10 single samples at 70 days interval were randomly sampled. Soil temperatures by digital geothermometer at sampling point time were measured. Pluviometric precipitation annual environmental temperature during the research were also quantified. Soil temperature ranging from 21.93 to 27.69 C at pinus area as well as from 22.22 to 26.58 oC at Corymbia one was reported. The monthly minimum relative humidity was 27.2/20.5 and maximum 82.6/63.2 as well as greatest precipitation levels for 22 days in January was observed. In relation to mycobiota, an increasing colony unit formation (CFU) difference at highest temperature and relative humidity ranging from 12.8 to 58.2 and 20.3 to 83.3 x 103 for Corymbia and pinus was reported. Fungi records total number presented 190 isolated ones from five genera and 54 different species: 32 Penicillium spp., 19 Aspergillus spp., one Eupenicillium javanicu, one Eurotium chevalieri and one Sclerocleista ornate, at all. The most abundant species was Penicillium decumbens, found in all samples. The periods of greatest concentration of CFU are correlated with periods of rain, humidity and temperature. Climatic variables as precipitation and temperature have been the elements which influenced the soil mycobiota changing the most.
